Pharmacokinetics and bioavailability of terazosin capsule in healthy volunteers

Objective:Pharmacokinetics and bioavailability of terazosin capsules and tablets were compared in 9 healthy volunteers after single-dose oral adminstration.Methods:The blood levels of terazosin were determined by RP-HPLC.The 3P87 program was used for the calculation of pharmacokinetic parameters.Results:Rate constants Ka were 8.2±4.0 and 6.4±0.4 h-1,rate constants T1/2 were 8.2±2.5 and 7.4±2.1 h.Peak plasma levels of capsules and tablets reached 43.5±8.5 and 43.1±0.8 ng.ml-1 ,at 0.61±0.11 and 0.94±0.39 h respectively.The AUC were 367.4±34.6 and 371.3±44.4 ng.h.ml-1 respectively.Conclusion:The pharmacokinetic parameters obtained from the study showed no significantly difference between two formulations (P>0.05).The relative bioavailability of capsules was 99.88% compared with the tablet.

The bioavailability of calcium carbonate effervescent granules in healthy volunteers

Objective:To investigate the bioavailability of calcium carbonate effervescent granules in healthy volunteers.Methods:The urinary calcium were determined by atomic absorption spectrophotometry after a single oral dose of calcium carbonate effervescent granules and calcium carbonate tablet was given to 10 healthy volunteers.Results:The urinary calcium following calcium carbonate effervescent granules was significantly higher whether expressed as the total amount or as the increment above basal (fasting) excretion.Conclusion:The bioavailability of calcium carbonate effervescent granules was higher than calcium carbonate tablet.

Preparation of human liver microsomes and determination of cytochrome P450

Objective:To study activity of cytochrome P450 for human liver microsomes.Methods:Three samples of human liver microsomes were prepared by density gradient centrifugation and the activity of Cytochrome P450 was determined.Results:cytochrome P450 was 0.523±0.005 nmol.mg-1;cytochrome b5 was 0.285±0.025nmol.mg-1;Aminopyrine N-demethylase was 0.524±0.06 nmol.mg-1;Ethylmorphine N-demethylase was 0.98±0.08.Conclusion:The activity of cytochrome P450 was affected by the reasons,and differed from individual.In the clinical,the different situation of every patient should be considered.

Study on mechanism of hypertension related to r-HuEPO

Objective:To investigate the pathogenesis of r-HuEPO related hypertension.Methods:Change of nitrous oxide(NO) production and endothelin-1(ET-1) was observed in anemi patients with chronic renal failure(CRF) during hemodialysis(HD) and anemic rats with CRF after 16 weeks of receiving r-HuEPO treatment resulting in hypertension and control groups.The level of plasma NO metabolites (NO-2/NO-3) and ET-1 was determined .Then inducible NO syunthase mRNA (iNOSmRNA) was detected by RT-PCR in rats renal cortical and medullary.Results:Hct、BP and the levels of plasma ET-1 were significantly higher (P<0.01) ,but the levels of plasma NO were markedly lower (P<0.05).There was no change of those in control groups (P>0.05).iNOSmRNA level was obviously lower in CRF rats than that of control rats. Conclusion:The decreases of iNOSmRNA and NO,and increase of ET-1 might play an important role in the development of hypertension related r-HuEPO.

Preparation of enteric thymosin tablets

Objective:To select the best formula for enteric thymosin tablets.Methods:The formula was orthogonal experiment used .The coating solution was composed of eudragit Ⅱ 4g,eudragit Ⅲ 2g,castor oil 2g,diethy1 phthalate 2.5g,tween-80 2g and 95% alcohol in 100ml.Results:The release rates in 0.1 mol.L-1 HCl solution (120min)and pH6.8 phosphatic buffer solution (45min) were 3.3%±0.9% and 93.6%±3.8%.Conclusion:The formula was simple.The coating tablets accorded with the rule in the CP.

Determination of fluorouracil and dyclonine hydrochloride Qu Bai Ling by dual Wavelength spectrophotometry

Objective:To establish a method of determination,concentration of fluorouracili and dyclonine hydrochloride in Qu Bai Ling.Methods:A dual wavelength spectrophotometry was used.The sample wavelength was 265 nm and the reference wavelength was 295nm for fluorouracili,the sample wavelength was 282nm and the reference wavelength was 245nm for dyclonine hydrochloride.Results:The correlation coefficients were both 0.9999.The average recoveries were 100.1% and 100.6%,with RSD 1.4% and 1.3% respectively.Conclusion:This method is sensitive rapid and accurate.

Determination of amoxicillin and clavulanate acid in amoxicillin and clavulanate potassium for oral suspension by HPLC

Objective:To establish a HPLC method for determination of amoxcillin and clavulanate acid in amoxicillin and clavulanate potassium oral suspension. Methods:A 3×3 CRC 18 column was used. The mobile phase was consisted of pH4.4 monosodium Phosphate buffle and methanol(95∶5).The determination wavelength was 220nm. Results:A linear concentration range of 25-500 μg.ml-1 Amoxicillin and 10～200μg.ml-1 clavulanate acid had been got.The relative recovery was 99.4%±1.9% and 99.5%±2.0%.The precision withday was 1.1%～2.3% and 1.7%～2.6%,the precision betweenday was <3.1% and <2.5%,respectively.Conclusion:The method is suitable for determination of the preparation.

Determination of the contents of total phosphorus of Cornu Antelopis and Stuff Antelopis and their water extract

Objective:To analysis the contents of total phosphoursin in Cornu Antelopis and Stuff Antelopis.Methods:The content of phosphorus was determined.Results:The total phosphorus in Counu Antelopis and its water extration was 0.179% and 9.19%,in stuff Antelopis and its water extract was 19.4% and 0.098%. Conclusion:The content of total phosphorus of Stuff Antelopis is higher than Cornu Antelopis's and Cornu Antelopis is far higher than stuff Antelopis in water-extract.

Study on the compatible stability of netilmicin with For Kinds Hemostatic

Antimicrobial test in vitro,Ultraviolet Spectrometry etc were used for the determination of Netilmicin with etamsylate,Aminomethylbenzoic Acid,Adrenosem and Aminocaproic Acid in 0.9% sodium chloride injection.The result showed that there was no evident variation in appearance,pH and content of all kinds ingredient at room tempreture.
