TGF-β1对瘢痕成纤维细胞α-SMA表达的诱导作用

    潍坊医学院000114摘 要:目的研究转化生长因子β1(TGF-β1)对瘢痕成肌纤维细胞中α-平滑肌肌动蛋白(α-SMA)表达的诱导作用。方法以5例增生性瘢痕为标本,3例正常瘢痕为对照,采用成纤维细胞二维培养和三维培养体系及LSAB免疫组织化学染色技术,观察了在不同TGF-β1浓度条件作用下,来源于增生性瘢痕和正常瘢痕的成纤维细胞表达α-SMA的情况。结果①不同浓度的TGF-β1诱导瘢痕成纤维细胞表达α-SMA的作用不同,以5ug/L的TGF-β1作用最有效;②与来源于正常瘢痕的成纤维细胞相比,来源于增生性瘢痕的成纤维细胞在表达α-SMA方面,对TGF-β1的反应更为敏感。③对同一瘢痕来源的成纤维细胞而言,三维培养体系中α-SMA的含量要明显低于二维培养体系。结论①在体外,TGF-β1诱导α-SMA在瘢痕成肌纤维细胞中的表达作用存在着浓度差异;②增生性瘢痕中成纤维细胞与正常瘢痕中成纤维细胞性状存在着差异,对TGF-β1敏感性不同;③细胞外基质成分可能会降低TGF-β1的诱导作用,使α-SMA的表达减少。

    The Induced Action by TGF-β1 in the Expression of α-SMA in Scar Fibroblasts

    Abstract:Objective To explore the expression of α-SMA actin induced by TGF-β1 in scar myofibroblasts and the responses of fibroblasts and the responses of fibroblasts from various scar origins in the course of the experiment.Method Five simples of hypertrophic scars and three samples of normal mature scars were collected as the experimental and control group respectively.The means of 2D and 3D fibroblasts cultured system and immunohistochemical method of LSAB were used to investigat the expression of α-SMA actin in myofibroblasts from hypertrophic scars and normal mature scars in the different conditions of TGF-β1 concentration.Result ①The expression of α-SMA action in myofibroblasts were different in response to various TGF-βl concentration.The reaction of TGF-β1 at 5 ug/L seemed to be more effective;②The fibrolasts from hypertrophic scars seemed to be more sensitive to TGF-β1,compared to the normal mature scars.③The contents of α-SMA in 3 dimension fibroblasts cultured system were markly lower than those in 2 dimension fibroblasts cultured system with respect to the same scar sources of fibroblasts.Conclusion ①The expression of α-SMA induced by TGF -β1 in scar myofibroblasts in vitro existed concentration discrepancy during the course of this experiment;②The qualities of the fibroblasts from hypertrophic scars and normal mature scars and their sensitivity to the inducement of TGF-β1 were different;③The inducement of TGF-β1 may be reduced by ECM compontents and that could cause the expression of α-SMA decreased.
