Hp cagA gene expression and application of its product

Abstract
AIM To express the cagA gene and develop a rapid immuno-detecting method for the infection of cagA+ Hp.
METHODS We cloned the 5-end of cagA (854bp) into the expression vector pBV220 and transformed DH5α with the plasmid pBV220/fcagA, in which a single CagA fragment (FCagA) was produced when the temperature reached 42℃. After being renaturated, FcagA was purified by anion exchange and sephadex G-100 chromatography. The FcagA had a relative molecular weight of 38000. With the prepared FCagA, coloidal gold, and immunogold, we established the dot immunogold filtratoin assay (DIGFA) to detect anti-CagA antibody in serum. ......
