Abstract
AIM To explore a culturing method of porcine hypatocytes on microcarriers and intend to use those culturing cells in bioartificial liver.
METHODS Hepatocytes were isolated from 1-2 months iocal pig by the two-step collagenase perfusion technique. 5×107 hepatocytes, 2g/L cytodex-3 microcarriers and 50mL DMEM including 100mL/L serum from porcine portal vein were inoculated into a 100mL flask. The morphology of culturing hepatocytes and concentrations of aibumin and urea in the supernatant were dynamically examined. ......
