灵芝治疗小鼠微小残留白血病的效果及其机制

     【摘要】 目的 探讨灵芝对微小残留白血病小鼠的治疗效果及作用机制。方法 采用微小残留白血病(MRL)小鼠模型,用药组灌胃灵芝0.4g/kg,每日1次,连续12或22天,观察小鼠生存期、白血病细胞浸润脏器(脾脏和骨髓)的病理变化,应用流式细胞仪检测白血病细胞凋亡及细胞周期改变,采用ELISA方法检测小鼠血清中TNF-α含量,同时测定外周血白细胞、淋巴细胞数及亚群、骨髓粒系祖细胞集落形成单位(CFU-G)能力,与MRL模型组和正常对照组相比较。结果 灵芝给药组小鼠生存期较MRL模型组明显延长(P<0.001)。给予灵芝12天后,浸润脾脏的白血病细胞凋亡明显增加、活跃的DNA合成被有效抑制,脾脏白血病细胞浸润减少,外周血肿瘤坏死因子(TNF-α)水平显著升高。而外周血白细胞和淋巴细胞数在治疗后第3、7天明显高于MRL模型组,骨髓CFU-G数明显增加,紊乱的T细胞亚群基本恢复,脾脏白髓数增加且白髓体积增大,提示小鼠体内免疫反应增强。结论 灵芝可通过诱导白血病细胞凋亡、促进造血和提高免疫功能,延缓残留白血病的复发,其机制与体内TNF-α水平显著升高、免疫功能增加密切相关。
