LFP 91-3 对小鼠腹水瘤细胞端粒酶及腹水碱性磷酸酶、胆碱酯酶活性影响

     【摘要】 目的 观察LFP 91-3 (香菇C 91-3 菌丝发酵液提取蛋白))对肿瘤细胞端粒酶及腹水中碱性磷酸酶、胆碱酯酶活性变化的影响。方法 纯系BALB/C小鼠腹腔注射H22肿瘤细胞造腹水瘤模型,然后腹腔注射LFP 91-3 抑瘤治疗,TRAP-PCR-ELISA法测肿瘤细胞端粒酶活性,自动生化分析仪连续监测法测定腹水中碱性磷酸酶、胆碱酯酶活性。结果 LFP 91-3 各治疗组肿瘤细胞端粒酶活性下调,且与LFP 91-3 的浓度成正比,与盐水对照治疗组比较P<0.01;LFP 91-3 50μg组、LFP 91-3 100μg组碱性磷酸酶活性较盐水对照治疗组降低(P<0.05);LFP 91-3 100μg组胆碱酯酶活性高于阳性抗肿瘤药物环磷酰胺组(P<0.01)。结论 LFP 91-3 能抑制肿瘤细胞增殖,使端粒酶活性降低是其抗肿瘤作用机制之一。在抗肿瘤作用的同时,LFP 91-3 毒性低于环磷酰胺。

    【Abstract】 Objective To investigate the effects of LFP 91-3 (lentinusdodes C 91-3 mycelium’s fermentative liquid protein)on telomerase activity of ascitic tumor cells and ascitic ALP,CHE activity.Methods Inbred BALB/C micewereinoculated with H22tumor strain to set up a model of ascitic tumor,and then injected with LFP 91-3 .The telomerase activity of ascitic tumor was analyzed quantitatively by TRAP-PCR-ELISA assay,and activity of CHE and ALP in ascites was detected by colorimetric method.Results Telomerase activity began to be down-regulated or inhibited when the tumor cells were treated with LFP 91-3 ......
