P16基因甲基化的检测及其在大肠癌诊断中的应用研究

【摘要】 目的 探讨P16基因甲基化与大肠癌发生发展的关系,研究从血清中筛选,诊断大肠癌的可行性。方法 采用MsP法检测36例大肠癌组织、7例大肠腺瘤瘤体组织及对应患者血清中P16基因甲基化状况,同时分析与临床病理特征的关系,10例健康志愿者血清标本作为对照组。结果 36例癌组织中检出P16基因甲基化15例,阳性率41.7%;癌组织标本中检出P16基因甲基化的15例对应患者血清标本中有9例也检测到同样改变,阳性率60%(9/15)。其余21例癌组织标本未检出P16基因甲基化其对应患者血清、7例腺瘤组织标本及对应患者血清,10例健康志愿者血清标本中均未检测到P16基因甲基化畸变。结论 P16基因甲基化与大肠癌的发生发展有关,血清中检测其变化可望成为监测患者预后的无创性诊断方法。关键词 P16基因 甲基化 大肠癌 血清
