肝癌放射治疗癌细胞辐射敏感性的预测研究

     【摘要】 目的 利用早熟染色体凝集技术预测γ射线对肝癌细胞辐射效应。 方法 用 60 Co产生的γ射线照射人肝癌细胞系SMMC-7721,以克隆形成法测定经照射后的细胞存活率,用化学诱导剂Calyculin-A.诱导的早熟染色体凝集技术研究染色体损伤。 结果 G2期细胞染色单体和等点染色单体断裂数与照射剂量之间存在着线性相关性;染色单体断裂总数与细胞存活率之间存在良好的线性相关性。 结论 辐射诱导的染色单体断裂是预测肝癌细胞内在辐射敏感性的一个良好指标,可为肝癌的临床放射治疗计划提供依据。

    【Abstract】 Objective To predictive the irradiation response of hepatoma cells exposed toγrays by using the prematurely chromosome condensation technique.Methods Hepatoma cell line SMMC-7721was exposed toγrays,cell survival fraction was measured by colony assay and chromosome damage was detected by chemically induced pre-maturely chromosome condensation echnique.Results Chromatid-type and isochromatid-type breaks have the lin-ear relationship in G2phase;the totalnumber of chromatid breaks was well correlated with the cell survival fraction.Conclusion This study implied that radiation-induced chromatid/isochromatid breaks can be possibly used as a good predictor of radiosensitivity of hepatoma cells ......
