Abstract:Allogeneic stem cell transplantation (SCT) is a standard therapy for hematological malignancies. Stable immunotolerance formation is the base for the successful transplantation. An important benefit of allogeneic SCT is the graft-versus-leukemia (GVL) effect. However, GVL effects are closely linked with grate-versus-host disease at present, a major cause of morbidity and mortality after allogeneic SCT. With the improved understanding of molecular mechanism for all those phenomena, it is possible to separate GVL from GVHD and improve the overall survival of allogeneic SCT for hematological malignancy. New and innovative immunotolerance methods will be developed in the near future and open a new era for mismatched allogeneic SCT.
