摘要 目的 观察腺病毒载体介导的人野生型Rb基因导入后促进血管平滑肌细胞衰老的作用,并探讨Rb基因抑制平滑肌细胞增生的机理。 方法 用外源性Rb基因重组腺病毒载体感染体外培养的兔主动脉血管平滑肌细胞。以细胞超微结构、3 H-胸腺嘧啶核苷掺入、流式细胞术分析细胞周期和β-半乳糖苷酶染色等方法观察平滑肌细胞的衰老变化。 结果 Rb基因导入可使平滑肌细胞发生衰老,表现为细胞线粒体肿胀,脂褐素堆积,DNA合成减少,细胞停滞在G0 /G1 期,与衰老相关的β-半乳糖苷酶表达增加。

结论 野生型Rb基因通过促进细胞衰老而对平滑肌细胞增生起抑制作用。

Promotion ofthe senescence of vascular smooth muscle cells by the introduction of retinoblastoma gene

Abstract Objective To observethe promotion of the senescence of vascular smooth muscle cells (SMC) after theintroduction of retinoblastoma (Rb) gene via a recombinant adenovirus vector and toexplore the mechanism of inhibition of SMC proliferation by wild-type Rb gene. Methods Areplication-deficient adenovirus vector encoding a wild-type Rb was constructed to infectthe cultured rabbit aortic SMC. The senescent changes of SMC were observed withtransmission electron microscope, 3 H-thymidineincorporation test, β-galactosidase stain and cellcycle analysis with flow cytometry. Results Introduction of wild-type Rb gene into SMC could arrest thecells at G0 /G1 phase of the cell cycle and lead to inhibition of DNAsynthesis.The senescent SMC showed swelling of mitochondria, accumulation of lipofuscinand positive senescence-associated β-galactosidasestain. Conclusions The results demonstrate that the wild-type Rb gene suppresses SMCproliferation by promotion of cell senescence.
