摘要 目的 研究人胰岛素样生长因子Ⅱ(IGF-Ⅱ)反义RNA对肝癌细胞株SMMC-7721的抑制效应。方法 人IGF-Ⅱ cDNA 0.1kb反向插入真核细胞表达载体pcDNA3,获得IGF-Ⅱ反义RNA表达载体pIGF-ⅡAs,将其导入人肝癌细胞株SMMC-7721,观察软琼脂培养集落形成的能力。流式细胞仪(FCM)测定pIGF-ⅡAs表达IGF-Ⅱ反义RNA对SMMC-7721细胞生长的影响。结果 转导入pIGF-ⅡAs的SMMC-7721细胞,不能在软琼脂上形成集落,FCM证实导入pIGF-ⅡAs细胞S期增多,而对照组SMMC-7721细胞和带pcDNA3空载体的SMMC-7721细胞则无明显变化。结论 pIGF-ⅡAs反义RNA体外可抑制肝癌细胞株SMMC-7721的致瘤性。

    

Inhibitory effect of IGF-Ⅱantisense RNA on malignant phenotype of hepatocellular carcinoma

Abstract Objective Inhibitory effect of insulin like growth factor Ⅱ(IGF-Ⅱ) antisense RNA on malignantphenotype of hepatic cancer cells was studied.Methods A 0.1kb cDNA ofhuman IGF-Ⅱ was reversely inserted into a eukaryotic expression vector of pcDNA3 and an IGF-Ⅱ antisense RNA expression vector of pIGF-Ⅱ As was obtained.The pIGF-Ⅱ As wasthen introduced into human hepatic cancer cell line SMMC-7721.The effect of IGF-Ⅱantisense RNA which was expressed by pIGF-ⅡAs on SMMC-7721 cell cycles progression wasmeasured with FCM.Results The coloning formation in anchorage-independentassay of SMMC-7721 cells with pIGF-ⅡAs was significantly decreased by comparision withcontrol groups of SMMC-7721 cells and SMMC-7721 cells transfected with pcDNA3 vectoralone.FCM analysis showed that the S phase of cell cycles for SMMC-7721 cells with pIGF-ⅡAswas increased,but there were no obvious changes in the control groups of SMMC-7721 cellsand SMMC-7721 cells with pcDNA3.Conclusion The pIGF-ⅡAs of IGF-Ⅱantisense RNA acts as an inhibitor on carcinogenesis of the SMMC-7721 cells in vitro.
