【摘要】 目的 初步研究庚型肝炎病毒NS3 蛋白片段在大肠杆菌中的表达及其免疫反应性。 方法 采用RT-PCR和基因重组技术从中国重庆市的庚型肝炎病毒(HGV)感染者血清中分离出HGV-NS3 区基因,定向克隆到质粒pQE-30中,构建高效表达质粒pQE-NS3 ,并在大肠杆菌中表达重组NS3 蛋白。 结果 经ELISA鉴定,该重组蛋白能特异性地与血清中的NS3 特异性IgG型抗体结合。 结论 重组NS3 蛋白经进一步纯化后,可用于血清学检测。
EXPRESSION OF HGV-NS3 PROTEIN IN E.COLI AND THE PRELIMINARY STUDY ON THE IMMUNOLOGICAL REACTIVITY OF THE RECOMBINANT HGV-NS3 PROTEIN

【Abstract】 Objective To express HGV-NS3 protein in E.coli and to approach the possibility of production of immunological reagent for the serological diagnosis of HGV. Methods According to the announced nucleiotide sequence of HGV, two sets of primers were designed in NS3 region of HGV and were subjected to RT-nested PCR. By cloning the amplified NS3 into pQE-30, the expression plasmid pQE-NS3 has been constructed, and subsequently transformed into E.coli(SG13009) to express NS3 protein. The recombinant HGV NS3 protein was identified with ELISA. Results HGV NS3 Protein was successfully expressed in SG13009 with IPTG induction, and its immunological reactivity has been identified with ELISA. Conclusion Expression of HGV-NS3 protein in E.coli was proved to be successful, and the recombinant HGV NS3 protein could specifically bind its counter-antibody.
