【摘要】 目的 研究鼻咽癌细胞系SUNE中EB病毒潜伏膜蛋白1(LMP1)基因对上皮细胞生长特性的影响,探索LMP1在鼻咽癌发生中所起的作用。方法 用LMP1基因真核表达质粒转染人胚肾上皮细胞,检测LMP1的表达,观察细胞的生长状态、生长速度、在软琼脂中的集落形成能力及对裸鼠的致瘤能力。结果 被LMP1基因转染的细胞生长旺盛,丧失接触抑制,生长速度增快,在软琼脂中能够形成多个集落，并能在裸鼠体内成瘤。结论 LMP1基因能明显改变上皮细胞的生物学行为，促进细胞的生长、增殖和转化，使转染的上皮细胞获得肿瘤细胞的生长特征。

    【Abstract】 Objective To study the effect of EBV latent membrane protein(LMP1) gene isolated from human nasopharyngeal carcinoma cell line SUNE on the growth ofepithelial cells and the role of LMP1 in the genesis of nasopharyngeal carcinomas. Methods EBVLMP1 gene eukaryotic expression plasmid was transfected into human embryo kidney epitheialcells. Expression of LMP1 protein in the transfected cells was detected. Dynamic cellgrowth status, growth rate, ability of colony formation in soft agar and thetumorigenicity of the transfected cells in nude mice were studied. Results The LMP1 genetransfected cells grew vigorously and rapidly, lost contact inhibition, formed morecolonies in soft agar, developed tumors in nude mice. ConclusionLMP1 gene could alter thebiological behaviours of epithelial cells, promote cell proliferation and transformation,and endows the transfected cells with growth characteristics like tumor cells.
