摘要 目的 研究MAL基因在人食管癌中的表达下调状况。方法 采用Northern杂交和RT-PCR方法检测了MAL基因在41对配对的食管癌组织和相应癌旁食管粘膜中的表达;应用PCR和RT-PCR方法分析了MAL基因在3个食管癌细胞系EC109、EC8712和EC9706中的存在及其表达。结果 MAL基因的表达水平在92.7%(38/41)食管癌组织中明显低于同一患者的癌旁食管粘膜。同时,MAL基因在3个食管癌细胞系中未见表达。PCR分析显示,这3个食管癌细胞系中均存在被分析的MAL基因片段。结论 MAL基因表达下调是食管癌发生、发展过程中的一个频发事件。

MAL gene is down-regulatedsubstantially in human esophageal cancer

Abstract Objective Toinvestigate the status of expression of MAL gene in human esophageal cancer. Methods Expressionof MAL gene was analyzed by Northern blot and/or RT-PCR in 41 pairs of human esophagealcancer tissues and matched adjacent normal mucosa, and three human esophageal cancer celllines EC109, EC8712 and EC9706. MAL gene was analyzed by PCR in the three esophagealcancer cell lines.Results MAL gene expression was down-regulatedsubstantially or barely detectable in 93%(38/41) of human esophageal cancer tissues whileexpressed at high level in all matched adjacent normal esophageal mucosa. In all 3 humanesophageal cancer cell lines, expression of MAL gene was not detectable. PCR analysisshowed that the MAL gene fragment analyzed was intact in the three esophageal cancer celllines.Conclusion Down-regulation of MAL gene expression is of frequentoccurrence in human esophageal cancer.
