【摘要】 目的 探讨肝纤维化组织学量化诊断方法。方法 用二甲基亚硝氨(DMN)诱导SD大鼠肝纤维化模型,常规肝组织切片,Masson三色和苦味酸天狼红染色,苦味酸固绿-天狼红染色比色测定,图像定量处理和半定量计分分析及羟脯氨酸(Hyp)含量测定。结果 切片胶原比色测定、图像定量处理和半定量计分分析均与肝纤维化程度变化相一致,与肝组织Hyp含量具有良好的相关性,r分别为0.94、0.89和0.88,P均<0.01。结论 切片胶原测定、图像定量处理和半定量计分可作为确定肝纤维化程度的量化指标。

【Abstract】 Ojbective To discuss the histological quantitativemethod of hepatic fibrosis. Methods Dimethylnitrosamine (DMN) was used toproduce an SD rat model for hepatic fibrosis and the effects with histological techniqueand liver hydroxyproline (Hyp) content observed. Histology technique included HE stain,Masson stain, Picric acid-Sirus red stain, and Picric acid-Sirus red stain colorimetrictechnique. Then picture quantitative analysis technique and semi-quantitative scoringmethod was used to analyse the effect of colorimetric determination. Results There was better consistency between the change of hepatic fibrosis and the colorimetricdetermination of collagen content, picture quantitative determination, semi-quantitativescoring method. There was better correlationship between Hyp content and determiningeffects of these three methods. The coefficient "r" of the three effects were0.94, 0.89 and 0.88, respectively(P<0.01). Conclusion These resultssuggest that it is possible to use collagen content determination in slice, picturequantitative determination and semi-quantitative scoring for the quantitative diagnosisindices in hepatic fibrosis.
