庆大霉素对兔成骨细胞增殖及超微结构的影响

【摘要】目的 观察庆大霉素对兔成骨细胞增殖及超微结构的影响。方法 从出生2周的新西兰兔颅盖骨获取成骨细胞,在含不同浓度庆大霉素的DMEM培养液中培养24小时或72小时,采用四唑盐比色法观察庆大霉素对成骨细胞增殖的影响,并对在含不同浓度庆大霉素的DMEM培养液中培养96小时的成骨细胞进行透射电镜观察。结果 高浓度庆大霉素组(800μg/ml、1600μg/ml)光吸收值明显低于对照组(P<0.01),高浓度庆大霉素组超微结构表现为内质网扩张,胞浆内见大量的吞噬溶酶体。结论 高浓度庆大霉素明显抑制成骨细胞增殖,呈现一定的毒性作用。

    Effects of Gentamycin onProliferation and Ultrastructure of Primary Cultured Rabbit Osteoblasts

Abstract Objective Toobserve the effects of gentamycin on the proliferation and ultrastructure of primarycultured rabbit osteoblasts. Methods Osteoblasts isolated from the two- week newly born rabbits werecultured for 24 h or 72 h in DMEM media with different concentrations of gentamycin. MTTmethod was used to observe the effect of gentamycin on cell proliferation. Theultrastructural changes of cultured osteoblasts exposed to gentamycin for 96 h were alsoobserved under transmission electron microscope. Results The opticaldensity values of higher concentration gentamycin groups (800μ g/ml, 1 600μ g/ml) were much smaller than that of the control group(P<0.01). The swelling of mitochondria and the presence of abundant phagolysosomes werecharacterized as the ultrastructural features. Conclusion Highconcentration gentamycin demonstrated a certain toxic inhibitive effects to osteoblasts.
