【摘要】 目的 观察增殖细胞核抗原(PCNA)基因反义RNA表达质粒对人胃癌细胞生长特性的影响,探讨其抗肿瘤作用。方法 用DNA重组方法将人PCNA基因反向克隆到真核表达质粒pDOR-neo中,构建成人PCNA基因真核表达质粒pDR-PCNA,用脂质体介导转染人胃癌细胞系SGC-7901。结果 经G418筛选获得转染细胞SGC/PCNA,与亲本细胞相比,其生长速度减慢,RNA、蛋白质的生物合成受到抑制,S期、G2 /M期DNA含量降低。结论 PCNA基因反义RNA对胃癌细胞的生长具有明显的抑制作用。

【Abstract】 Objective To observe the effect of proliferating cell nuclearantigen (PCNA) antisense RNA on the growth characteristics of human gastric cancer cell.Methods An eukaryotic expression vector pDR-PCNA with human PCNA gene insert in reversedirection was successfully constructed. The human gastric cancer cell line SGC-7901 wastransfected with pDR-PCNA by lipofectamineTM . Results Growth rate, proteinand RNA biosynthesis and DNA contents in S-phase and G2 /M-phase of thetransfected SGC-7901 were significantly suppressed in comparison with those of theparental cell line. Conclusion PCNA antisense RNA can significantly suppress the invitro growth of gastric cancer cell line.
