【摘要】 目的 观察钙网蛋白(CRT)在瘢痕及正常皮肤成纤维细胞内的表达情况。方法 采用免疫细胞化学染色和原位ELISA技术对体外培养的瘢痕及正常皮肤成纤维细胞进行CRT分布定位及表达水平研究。结果 瘢痕和正常皮肤成纤维细胞在处于增殖生长状态时,胞浆及核内均有不同程度的CRT表达,其中瘢痕成纤维细胞表达CRT水平(A:1.97±0.15,ELISA492nm)明显高于正常皮肤成纤维细胞水平(A:1.43±0.12),差异有非常显著意义(P<0.01);而在已融合的瘢痕和正常皮肤成纤维细胞内则均无CRT表达。结论 瘢痕成纤维细胞内CRT的丰富表达对促进细胞迁移、信号传递及钙存储等方面均有着积极作用。
Study on the expression ofcalreticulin in hypertrophic scar-derived fibroblasts

【 Abstract 】 Objective Toinvestigate the expression level and intracellular distribution of calreticulin(CRT) inhypertrophic scar-derived fibroblasts(HSF)and normal skin-derived fibroblasts(NSF).Methods Using anti-CRT antibody, CRT was detected in HSF and NSF by immunocytochemistry and ELISAtechnique.Results CRT was present in the cytoplasm and nuclei inproliferating HSF or NSF in culture, but HSF possessed significantly higher expressionlevel of CRT than NSF(P<0.01). When these cells (HSF and NSF) were fused , neitherintracellular nor intranuclear staining for CRT was observed.Conclusion In HSF and NSF,the distribution of CRT is widespread. Because of its abundant expressionin HSF, we postulate that CRT as a multifunctional protein plays an important role in Ca2+ sequestering, integrin-mediated signalling and cell adhesion.
