ahpC基因突变与结核分支杆菌耐异烟肼的研究

【摘要 】 目的 了解我国结核分支杆菌耐异烟肼(INH)分离株ahpC编码基因及其启动子突变情况,研究其与INH耐药关系。方法 通过聚合酶链反应(PCR)-单链构象多态性(SSCP)方法分析62株结核分支杆菌临床分离株ahpC及其启动子基因。以H37 RV 标准株为对照。结果 32株结核分支杆菌药物敏感株ahpC及其启动子基因SSCP均泳动正常;30株耐INH分离株ahpC编码基因SSCP也未见异常;12株高度耐INH分离株中,5株ahpC启动子SSCP泳动异常;18株低度耐INH分离株的ahpC启动子泳动正常。结论 结核分支杆菌ahpC编码基因与耐INH无关;ahpC启动子突变是结核分支杆菌katG改变、过氧化氢酶缺乏的代偿性变化,也许能作为结核分支杆菌高度耐INH的间接指标。

Abstract 】 Objective To investigate the mutations ofahpC genes in M. tuberculosis isoniazid-resistant isolates, and to study the correlationbetween ahpC alterations and isoniazid resistance. Method Analyzing the ahpC codingsequences and ahpC promoters in 62 M. tuberculosis clinical isolates with PCR-SSCP method.M. tuberculosis strain H37 RV was used as control. Result The ahpC coding sequences showednormal SSCP profiles in 62 clinical isolates. AhpC promoter mutations were identified in 5of 12 high isoniazid-resistant strains, and were not altered in 18 low isoniazid-resistantand 32 drug-sensitive isolates. Conclusion The mutation of ahpC coding gene was not correlated with isoniazid-resistant M. tuberculosis isolates. AhpC promoter mutation was a compensatorychange in katG-defective and catalase-negative isolates, may be used as a indirect markerfor high resistance to isoniazid.......
