炎症显像剂99Tcm-citrate的制备及动物实验

【摘要】 目的 研制炎症显像剂99 Tcm -柠檬酸盐(citrate)。方法 柠檬酸三钠在还原剂氯化亚锡存在,pH值6.0时与99 Tcm 络合,生成99 Tcm -citrate。研究了标记物的稳定性、炎症模型小白鼠体内分布并进行了炎症模型大白兔显像。结果 制备的99 Tcm -citrate标记率大于90%,6h内稳定。小白鼠炎症组织与正常组织的放射性比值高,注射后5、30、60、120min分别为0.97、3.18、4.76和3.32。99 Tcm -citrate主要经肾脏排泄,血液清除快。兔显像表明:注射后30、60、120、240min,炎症部位与对侧正常组织放射性比值分别为2.30、2.68、3.40和3.31。结论 99 Tcm -citrate是一种有效灵敏的炎症灶显像剂,且制备方便。

【Abstract】 Objective Toprepare 99 Tcm -citrate, an inflammation tracer. Methods Using Sn(Ⅱ) as reducing agent, sodium citrate was complexed with 99 Tcm at pH 6.0. The stability of yielded complex, 99 Tcm -citrate andits biodistribution in mice model were studied.99 Tcm -citratescintigraphy in rabbit model of inflammation was performed. Results Thelabelling rate was over 90%.99 Tcm -citrate was stable within 6hours at room temperature. The inflammatory tissue/normal tissue were 0.97, 3.18, 4.76 and3.32 at 5, 30, 60 and 120min after injection, respectively. The radioactivity clearancemainly occurred in the kidney, and the clearance from blood was rapid. Images acquiredfrom the rabbit model showed that the inflammatory/normal radioactivity ratio were 2.30,2.68, 3.40 and 3.31 at 30, 60, 120 and 240 min, respectively. Conclusions 99 Tcm -citrateis an effective and sensitive radiopharmaceutical for scintigraphy of inflammatorylesions, and it is easy to prepare.
