【摘要】目的评价顺铂对原代胃癌细胞的体内外杀伤效应。方法采用纯化后的人体新鲜胃癌细胞，用台盼蓝染色法和四甲基偶氮唑蓝(MTT)法检测经顺铂作用后细胞活力及代谢活性的变化；另用末端脱氧核苷酸转移酶法(TdT法)和正常免疫小鼠肾包膜下种植法(SRC法)分别检测顺铂体外促凋亡效应及体内抑瘤率。结果经顺铂作用后，肿瘤细胞出现明显的形态学改变和代谢活性的下降；不同个体肿瘤细胞对顺铂敏感性不同，低分化胃癌细胞较高分化者更敏感(40.25%比34.26%)；10倍血药浓度下顺铂对肿瘤细胞的抑制率高于单倍浓度(40.10%比38.45%)。经顺铂作用后，原代胃癌细胞凋亡率增加，为32.12%。顺铂还能明显抑制体内移植瘤的生长。结论顺铂可能通过包括诱导凋亡在内的多种途径杀伤原代胃癌细胞，尤对低分化者显示了明显的肿瘤杀伤效应。
Antitumor effect of cisplatinin vivo and in vitro on fresh human gastric cancer

【 Abstract】 Objective To evaluate the antitumor effect ofcisplatin in vivo and in vitro on fresh human gastric cancer cells. Methods Highlypurified gastric cancer cells were obtained from 50 resected primary lesions for the MTTassay and trypan blue exclusive assay. The degree of apoptosis induced by cisplatin wasdetermined by terminal deoxynucleotidyl transferase(TdT) assay in 20 cases.Simultaneously, the inhibitory effect of CDDP in vivo was investigated by subrenal capsuleossay(SRC)assay in 24 cases . Results Remarked morphologic and metabolic changes , whichvaried from one patient to another,were found in gastric cancer cells after exposure to cisplatin. Poorly differentiated gastric cancers were more sensitive to cisplatin thanwell differentiated ones (40.25% vs 34.26% ). The inhibition rate increased with theelevation of drug concentration. The average apoptic rate induced by CDDP was 32.12% infresh human gastric cancer cells. The obvious inhibitory effect of CDDP in vivo was alsoshowed by SRC assay. Conclusions A complex mechanism may be used to explain the antitumoreffect of CDDP, which might be a good candidate in chemotherapy for poorly differentiatedgastric cancers.
