     摘要 目的 探讨雄激素对成骨细胞代谢的影响。方法 胎鼠头盖骨成骨细

    胞培养于含不同浓度睾酮的培养基中,观察细胞胸腺嘧啶核苷掺入、碱性磷酸酶(ALP)活性、骨钙素基因表达及骨钙素合成等细胞增殖及分化等指标的变化情况。结果 胎鼠头盖骨成骨细胞的增殖不受睾酮影响,但ALP活性、骨钙素基因表达及其合成在不同程度上受睾酮的调节。结论 雄激素对成骨细胞的分化具有促进作用,但对其增殖无影响。

Effects of testosterone onfetal rat calvarial osteoblastic cells in vitro

AbstractObjective To investigate the effect of testosterone on bone cell metabolism.MethodsFetal rat calvarial osteoblastic cells were cultured in the medium containing differentconcentrations of testosterone.3 H-thymidine incorporation,alkaline phosphatase(ALP) activity,osteocalcin gene expression and production were detected todetermine the condition of cell proliferation and differentiation.ResultsThe proliferation of fetal rat calvarial osteoblastic cells was independent of thepresence of testosterone.However,ALP activity,osteocalcin gene expression and productionwere regulated in different extent by testosterone. ConclusionTestosterone stimulates the differentiation of rat calvarial osteoblastic cells in vitro.
