     摘要:目的:建立珍珠梅中槲皮素反相高效液相色谱法(RP-HPLC)含量测定方法。方法:采用RP-HPLC对槲皮素进行定量测定;采用YMC-pack ODS-A(4.6 mm×250 mm,5 μm)色谱柱,检测波长为370 nm。结果:色谱条件为流动相甲醇0.2%磷酸溶液(50∶50),柱温为35℃,流速0.4 ml·min1。此法线性范围0.025~0.15 μg,r=0.999 9,精密度(RSD)为0.72%(n=5);平均回收率为98.8%,RSD为0.69%(n=5)。结论:测定方法简便快捷、精密度高、准确性好,可以作为珍珠梅中黄酮的测定方法,以控制其原料的质量。

    Determination of Quercetin in Sorbaria sorbifolia by RP-HPLC Assay

    Abstract:Objective:To establish an RP-HPLC method for the determination of quercetin in Sorbaria sorbifolia.Methods:The content of quercetin was determined by RP-HPLC. YMC-pack ODS-A(4.6 mm×250 mm,5 μm)column was used. The detection wavelength was set at 370 nm.Results:The mobile phase consisted of methanol-0.2% phosphatic acid (50∶50) with flow rate 0.4 ml·min1 and column temperature at 35℃. The method was linear within the range of 0.025~0.15 μg,(r=0.999 9). The method RSD was 0.72%(n=5). The average recovery of assay was 98.8% and RSD 1.45%(n=5).Conclusion:The method is simple, rapid and highly accurate and precise. It can be used for the quantitative determination of quercetinin Sorbaria sorbifolia. ......
