促红细胞生成素对缺氧缺血新生大鼠脑水肿和NO、MDA含量的影响

     【摘要】 目的 探讨重组人红细胞生成素(rhEPO)对新生大鼠缺氧缺血性脑损伤(HIBD)、脑水肿及NO、MDA含量的影响。方法 选用7日龄Wistar大鼠80只随机分为4组,即假手术组、缺氧缺血性脑病组(即生理盐水对照组)、促红细胞生成素(rhEPO)治疗组、硫酸镁治疗组,每组各20只。后3组制备HIBD模型前30min分别腹腔内注射生理盐水、rhEPO及硫酸镁。4组动物48h后处死,随即取鼠分别给予4种处理:(1)取脑组织常规固定、切片、行H-E染色,观察脑组织的病理改变;(2)一组取脑组织精确称重后,置于恒温烤箱内48h后称干重求出脑的含水量;(3)取脑制成组织匀浆,测NO含量;(4)取脑制成组织匀浆,测MDA含量。结果 假手术组脑的含水量及NO和MDA含量均明显低于HIBD组,差异有显著性(P<0.05); rhEPO治疗组测定值明显低于生理盐水对照组(P<0.05),而与硫酸镁治疗组相比差异无显著性(P>0.05)。结论 促红细胞生成素(rhEPO)和硫酸镁一样对脑缺氧缺血具有保护作用,可以减轻脑水肿,抑制自由基的生成。

    Protective effects of rhEPO on hypoxic-ischemic brain damage in neonatal rats

    【Abstract】 Objective To observe the effects of recombinant human erythropoietin (rhEPO) on the cerebral edema and the contents of NO, MDA of the hypoxic-ischemic rat brain. Methods 80 infant Wister rats of 7 days' old were distributed randomly into 4 groups with 20 infant rats for each group: sham-operated control group, hypoxic-ischemic brain damage (HIBD) group, rhEPO treated group and MgSO4 treated group. Each rat in the last 3 group was killed at 48 hours after HIBD. rats were randomly taken out from each group to observe the pathologic section, and to measure the contents of water, NO and MDA. Results The levels of water contents, NO and MDA in the sham-operated control group were all lower than that in the HIBD group (P<0.05). And the levels in the rhEPO group were lower than that in the HIBD group (P<0.01). There was no difference in the levels between the rhEPO group and the MgSO4 treated group(P>0.05).Conclusion rhEPO can protect the brain tissue of HIBD, relieve the cerebral edema, and inhibit the production free radical. ......
