毛细管区带电泳分离和检测利多卡因方法的研究

     【摘要】 目的 建立毛细管电泳分离和检测利多卡因的方法。方法 毛细管区带电泳分离利多卡因,电泳分离条件:30mmol/L NaH2PO4-H3PO4(pH=5.0)缓冲液。先采用压力(20psi)进水5s,随后电动进样(8kV,5s),检测波长200nm,运行电压30kV,温度20℃,运行时间5min。结果 在0.1~12.5μg/ml范围内呈良好的线性关系和重现性(R2=0.9998,n=5),回收率96.80%~105.8%,日内及日间精密度(RSD)均小于6%。方法的检测限10ng/ml。结论 本方法简便、快速、灵敏,为利多卡因的定量测定提供了新的、更为理想的方法学手段。

    Determination of lidocaine by capillary zone electrophoresis

    【Abstract】 Objective To establish capillary zone electrophoresis method for the determination and separation of lidocaine.Methods Determination and separation of lidocaine by capillary zone electrophoresis (CZE) and electrophoresis conditions were as following: buffer was 30mmol/L NaH2PO4-H3PO4 solution (pH=5.0), the working voltage at 30kV, temperature at 20℃, UV detection at 200nm, sampling injected 5 seconds with voltage at 8kV.Results The established method had a good linearity and precision within the concentration range of 0.1~12.5μg/ml(R2=0.9998). The range of recovery was 96.8%~105.8%, and relative standard deviations of inter-day and intra-day were less than 6%. The detection limit was 10ng/ml.Conclusion This assay was simple, rapid and sensitive. It is a new and ideal analytical means for the determination of lidocaine. ......
