摘要: 目的: 比较两种不同方法检测急性白血病(AL)患者Bcl2和Bcl2/Bax表达情况。方法:采用链亲和素胶体金原位杂交法(ISHSAG)和间接免疫荧光法对57例AL患者的Bcl2和Bcl2/Bax进行检测。结果: (1) ISHSAG法中,完全缓解(CR)组Bcl2细胞表达率高于间接免疫荧光法,且差异有统计学意义(P<0.05)。(2) Bcl2及Bcl2/Bax检测均显示ISHSAG法的特异度、准确度、阳性预测值、阴性预测值、阳性似然比、阴性似然比优于间接免疫荧光法;Bcl2/Bax检测也显示ISHSAG法较间接免疫荧光法高。(3)ISHSAG法中Bcl2细胞表达率和积分值的阳性检出率无差别。结论: (1) ISHSAG法测定Bcl2及Bcl2/Bax优于间接免疫荧光法。(2)ISHSAG法中Bcl2积分值临床应用价值不大。关键词: 间接免疫荧光; 原位杂交; Bcl2; Bcl2/Bax
Comparision between indirect immunofluorescence assay and in situ hybridization assay in detecting the expression of Bcl2 and Bcl2/Bax in acute leukemia CHEN Rong, LI Ling, WEN Bingzhao
(Department of Hematology, First Affiliated Hospital, Xinjiang Medical University,Urumqi 830000, China)Abstract: Objective: TO compare two different assays by detecting the expression of Bcl2 and Bcl2/Bax in acute leukemia(AL) cells.Methods: Fiflyseven AL patients were detected for the expression of Bcl2 and Bcl2/Bax by in situ hybridization with streptavidingold(ISHSAG) assay and indirect immunofluorescence assay. Results: (1)The cell expression rate of Bcl2 in those three groups detected by ISHSAG were all significantly higher than that by indirect immunofluorescence assay (P<0.05). The negative expression rate of Bcl2 in complete remession(CR) group showed the same result. As to Bcl2/Bax,there was no significant difference of approach in those three groups.
