大鼠胰膀胱引流式单纯胰腺移植模型的建立

【摘要】目的:建立大鼠胰膀胱引流式单纯胰腺移植的动物模型。方法:22只脲链霉素诱导的糖尿病SD大鼠行单纯胰腺移植,正常22只SD大鼠为供体;血管吻合采用供胰腹腹主动脉(含脾动脉)和门静脉袢(含脾静脉)与受体腹主动脉及左肾静脉分别行端侧和端端吻合(袖套式);外分泌系统采用胰膀胱引流式。于术前2d和术后1,3,7,14,30d监测受体体重、进食量、饮水量和空腹血糖,并分析失败原因。结果:供体平均手术时间(33.7±9.5)min,受体平均手术时间(49.4±13.4)min,移植物均无热缺血时间,冷缺血时间(44.4±15.8)min。除12例于1个月内死亡或胰腺失去功能外,均成活1个月以上。术后并发症主要为尿路并发症(7例,31.8%)。成功的单纯胰腺移植后受体第1日血糖即下降后基本稳定。结论:成功的单纯胰腺移植均可有效地恢复糖尿病大鼠内分泌功能。【关键词】胰腺移植·模型,动物·大鼠
The establishment of experimental model of rat pancreas transplantation alone with bladder drainage

【ABSTRACT】Objective:To establish the rat model of pancreas transplantation alone(PTA) with bladder drainage.Methods:22 streptozotocin induced diabetic rats received PTA from the normal donors.Venous drainage of PTA performed using endtoside anastomosis between the donors’ abdominal aorta segment and recipients’ abdominal aorta,and endtoend anastomosis between the donors’ portal vein segment and recipients’ left renal vein(use a cuff).Pancreas exocrine drainage was made by pancreas bladder anastomosis(RouxY).The blood glucose,body weight,food intake,water intake and urine volume were monitored before transplantation and on the 1st,3rd,7th,14th,30th day after it.Results:The mean duration of operation of group PTA in donors was (33.7±9.5) min,and that in recipients was(49.4±13.4) min.The mean duration of warm ischemic time was 0 min and that of cold ischemic time was (44.4±15.8) min.12 of the 22 cases in group PTA died or lost their function of the endocrine within 1 month.The main complications were followed urinary tract complications after transplantation (7 cases,31.8%).All successful recipients' blood glucose lowed on the first day after transplantation(P0.01) ......
