【摘要】目的:以转染HBV全基因组并稳定分泌病毒抗原的HepG2.2.15肝癌细胞系为细胞模型,探索IFNγ对HBV相关性肝癌的免疫调节效应。方法:用HBsAg 和HBeAg ELISA检测试剂盒测定IFNγ作用0,6,12,24h后,HBV病毒颗粒的分泌表达水平。通过MTT法检测不同剂量IFNγ对新型凋亡配体TRAIL与TWEAK的生长抑制效应的调节作用,不同剂量IFNγ对TRAIL与TWEAK的凋亡诱导效应的调节作用。结果:IFNγ在不同的时间点对HBsAg和HBeAg的分泌均具有抑制作用,IFNγ能够显著增强TRAIL和TWEAK对HepG2.2.15肝癌细胞系的生长抑制效应和凋亡率。讨论:IFNγ是HBV感染后的一个重要的免疫效应分子,它可以通过抑制病毒复制和增强免疫监视分子的效应而在HBV相关性肝癌中发挥抗病毒和抗肿瘤效应。【关键词】干扰素γ,重组·肝肿瘤·肝炎病毒,乙型·配体·细胞凋亡·免疫,细胞
Immune regulation effect of IFNγ on HBV related hepatocarcinoma

【ABSTRACT】Objective:The HBV transgenic HepG2.2.15 hepatocarcinoma cell line was used as a cell model to explore the immune regulation effect of IFNγ on the HBV related hepatocarcinoma.Methods:The HBV particles secreted by HepG2.2.15 cells were detected by HBsAg and HBeAg detecting ELISA kits.Modulation of IFNγ to the the growth inhibiton effect induced by apoptosis inducing ligand TRAIL and TWEAK were assayed by MTT method.Modulation of IFNγ to the apoptosis inducing effect of TRAIL and TWEAK were assayed by flow cytometry.Results: Secretion of HBsAg and HBeAg could be inhibited by IFNγ.The growth inhibition effect and apoptosis induced by TRAIL and TWEAK could be greatly increased by IFNγ.Conclusion:IFNγ is a very important immune effective molecule that will play a key role after HBV infection.It could exert its immune function by inhibiting the virus secretion and enhance the immune surveillance in HBV related hepatocarcinoma. ......
