高眼压视网膜缺血再灌注中脑组织Fos的表达及神经营养因子对其表达的影响

     摘要:目的: 观察视网膜缺血再灌注损伤过程中脑组织中Fos的变化及神经营养因子对Fos表达的影响,探讨Fos在脑组织不同部位的改变及可能作用机制。方法:建立大鼠高眼压眼缺血再灌注损伤动物模型,分为对照组及实验组Ⅰ、Ⅱ、Ⅲ组,实验Ⅰ组为缺血组(I组),实验Ⅱ组为缺血2 h+再灌注组(I/R组),实验Ⅲ组为缺血再灌注前于右侧侧脑室注射神经营养因子(神经营养因子+ I/R组)。采用HE常规染色切片,光镜下观察脑外侧膝状体、嘴侧丘、视皮质结构神经细胞变化,采用免疫组织化学染色分别测定各组大鼠以上3个部位Fos的变化。结果:(1)对照组、缺血组、缺血再灌注组大鼠脑组织外侧膝状体、嘴侧丘、视皮质间Fos蛋白的表达差异无统计学意义(P>0.05)。(2)再灌注组与缺血再灌注组相比,外侧膝状体中Fos的表达差异有统计学意义(P<0.01)。(3)再灌注1 d组与再灌注2 d组间差异有统计学意义。(4)再灌注后5 d组与2 d组间Fos差异无统计学意义(P>0.05)。(5)实验Ⅲ组脑组织中不同部位的Fos与对照组相比差异无统计学意义。结论:(1)Fos在脑外侧膝状体、嘴侧丘、视皮质参与损伤的作用。(2)外侧膝状体中Fos的变化在脑损伤的中枢调节中有一定作用。(3)神经营养因子能明显减轻脑损伤反应。

    The expression of Fos in brain following retina ischemiareperfusion injury in rats

    Abstract: Objective: To observe the changes of Fos in injuried brain following retina ischemiareperfusion injury, the effect of nerve nutritive factor on Fos and evaluate the contribution of Fos to brain tissue effect. Methods: A model of retina ischemiareperfusion injury was established by imitation. The contents of Fos in brain tissue including lateral geniculate body and visual area regions were examined individually by immunohistochemical technique following sham operation(group1), ischemia without reperfusion(group2), reperfusion following ischemia (group3), the last groups(group4) injected with NGF. Results: Compared with groups from 1 to 3, the contents of Fos in brain tissue including lateral geniculate body and visual area regions had obviously statistic significance(P<0.05). There was no changes between group1 and group4 (P>0.05). The increase exists remarkably about the the contents of Fos in lateral geniculate body compared with the others. while no changes happen between group8 and group7 (P>0.05). Conclusion: The data of Fos in brain tissue including lateral geniculate body and visual area regions vary depending on different time of retina ischemiareperfusion specially, after reperfusion. Fos are involved in brain tissue injury following reperfusion of ischemia retina. The result also suggested that NGF can have a positive effect on retina ischemiareperfusion injury. ......
