大鼠肾缺血再灌注后一氧化氮合酶在肾组织中的表达

     摘要:目的:观察大鼠肾缺血再灌注(I/R)后一氧化氮合酶(NOS)在时间和空间上的表达特点及肾组织肾单位中各部位对损伤的敏感程度,探讨一氧化氮(NO)在介导肾组织损伤中发挥的双重作用及肾缺血再灌注后治疗损伤肾组织的最佳时段。方法:建立大鼠肾缺血再灌注模型,SP免疫组化法检测不同时间段NOS的表达变化。结果:对照组NOS的表达极低, 在大鼠髓放线、肾小体中几乎不表达NOS。在近曲小管、远曲小管中,各组与对照组相比较NOS表达差异均有统计学意义(P<0.05),I/R 2 h、I/R6 h、I/R10 h、I/R14 h 4组组间相比较差异也具有统计学意义(P<0.05)。各组中NOS在I/R2 h开始表达,同时近曲小管较远曲小管NOS表达明显增高,I/R10 hNOS的表达至高峰, I/R14 hNOS表达呈轻度下降趋势。结论:NO在介导肾组织损伤中发挥双重作用,主要损伤部位是近曲小管,其次是远曲小管,肾小体、髓放线对损伤不敏感。

    Expression of NOS in the rats' kidneys after renal ichemia reperfusion

    Abstract: Objective: To investigate the changes of the expression of NOS in rats′ kidneys after ischemia/ reperfusion and their relations to renal injury. To discuss situation of renal injure and NO plays an important role in rats' kidneys after ischemia/ reperfusion. Methods: Renal ischemia/reperfusion injury models of rats were established and SP immunohistochemical methods were used to detact the changes expression of NOS. Renal ischemia/reperfusion injury were also observed. Results: The expression of NOS increased in the kidneys ischemia/reperfusion gradually upregulated with the duration of ischemia/reperfusion and peaked at 10 h of reperfusion(P<0.05), then decreased slightly. The pathological changes and expression of NOS mainly occurred in renal proximal convoluted tubules. The second is in renal distal tubular convoluted tubules. NOS are hardly expressed. Conclusions: These results suggest that NOS may be involved in the process of renal ischemia/reperfusion in jury,the most seriously pathological changes are renal proximal convoluted tubules. The second is in renal distal tubular convoluted tubules.renial corpuscles are hardly influencend. ......
