癫痫发作大鼠模型行为学表现与脑组织学变化

     摘要:目的:观察匹罗卡品(Pilocarpine,PCPN)诱导的癫痫大鼠模型行为学表现与脑颞叶海马结构的组织学变化。方法:成年Wistar雄性大鼠33只随机分成3组:正常对照组3只,腹腔内注射1 ml生理盐水;实验Ⅰ组15只,腹腔内注射350 mg/kg PCPN,观察2 h;实验Ⅱ组15只,腹腔内注射380 mg/kg PCPN,观察15 d。光镜下观察海马齿状回和CA3区神经细胞变化。结果:(1)行为学癫痫发作行为表现遵循Racine标准Ⅰ~Ⅴ级循序渐进过程,实验Ⅰ组诱发成功率为57.1%,实验Ⅱ组诱发成功率为92.3%,两组相比差异有统计学意义(P<0.05)。癫痫发作诱导期时间随给药剂量不同而明显不同,实验Ⅰ组明显长于实验Ⅱ组(P<0.05)。(2)组织学变化:两实验组海马齿状回和CA3区以细胞坏死、水肿为主,未见胶质细胞增生。与正常对照组、实验Ⅱ组相比,实验Ⅰ组海马齿状回、CA3区的颗粒层神经细胞数量明显减少(P<0.05);实验Ⅱ组坏死区周围可见胶质细胞增生。结论:PCPN能诱导大鼠癫痫,脑颞叶海马神经细胞损伤程度随癫痫持续时间长短而不同。

    Analysis on behavioral and morphological alterations after seizures in rats

    Abstract:Objective: After seizures produced by pilocarpine in rats, the behavioral and morphological alterations of hippocampus were observed. Methods: Thirththree adult male Wistar rats were divided into three groups. One control groups; Experimental groups Ⅰ treated with pilocarpine in a dose of 350 mg/kg and observed 2 hours; Experimental groups Ⅱ with a dose of 380 mg/kg, observed 360 hours. All behavioral assessments were recorded according to Racine' criterion, normal nerve cells were counted under light microscope. Results: The induced time was different depending on the given dosage. The cell necrosis and endema were observed in dentate gyrus and CA3 in two experimental groups. The nerve cells were remarkablely decreased in experimental group Ⅰ; The microglia hyperplasia were observed in experimental group Ⅱ. Conclusion: The higher dose of pilocarpine, 350~380 mg/kg, can lead to seizures of rats. The morphological alterations depends on the lasting time of seizures. ......
