草棉花提取物聚Ⅰ和聚Ⅱ对ADP和胶原诱导的大鼠血小板聚集功能的影响

     摘要: 目的: 探讨草棉花提取物聚Ⅰ和聚Ⅱ对ADP和胶原诱导的大鼠血小板聚集功能的影响。 方法: 以二磷酸腺苷(Adenosine5′Diphosphate,ADP,终浓度为3.33 μmol/L)和胶原(Collagen,终浓度为212 μg/L)诱导血小板聚集,聚Ⅰ和聚Ⅱ分别分为25.0、50.0、100.0 mg/kg 3个剂量组,阿司匹林(17.1 mg/kg, ig)和丹参滴丸(72.9 mg/kg,ig)为阳性对照,记录5 min内聚集曲线,以最大聚集率和抑制率为观察指标。 结果: 聚Ⅰ(50.0、100.0 mg/kg, ig)、聚Ⅱ(50.0、100.0 mg/kg, ig)对ADP诱导的大鼠血小板聚集有明显抑制作用(P<0.05或0.01),抑制率分别为16.79%、21.98%和11.71%、17.67%,呈剂量依赖性。聚Ⅰ(100.0 mg/kg, ig)对Col诱导的血小板聚集有明显抑制作用(P<0.05),抑制率为13.35%。 结论: 聚Ⅰ对血小板聚集抑制作用较聚Ⅱ强,可在确定聚Ⅰ提取工艺的基础上进一步优化。

    The effects of Gossypium Herbaceum extract on platelet aggregation in rats

    Abstract: Objective: To explore the effects of Gossypium Herbaceum extract, Extract Ⅰ and Extract Ⅱ, prepared in two extraction methods, on platelet aggregation in rats induced by ADP and collagen. Methods: Extract Ⅰ and Extract Ⅱ were given at three doses (25、50、100 mg/kg). Aspirin (17.1 mg/kg, ig) and Salvia Miltiorrhiza(72.9 mg/kg,ig)were used as positive controls, ADP(Adenosine5′Diphosphate, final concentration was 3.33 μmol/L) and Col (Collagen, final concentration was 212 μg/L) were used to induce platelet aggregation. Turbidity method was used to assess the platelet aggregation function in rats. Maximum aggregation rate was observed as the index. Results: Extract Ⅰ(50、100 mg/kg, ig)、Extract Ⅱ(50、100 mg/kg, ig)had a marked inhibition effect on the ADPinduced platelet aggregation in rats (P<0.05 and P<0.01, respectively). The inhibition rates were 16.79%、 21.98% and 11.71%、 17.67%, demonstrating dosedependance. Extract Ⅰ(100 mg/kg, ig)also showed an inhition for Colinduced platelet aggregation. The inhibition rates was 13.35%. Conclusion: The antiplatelet aggregation effect of Extract Ⅰ is more potent than that Extract Ⅱ and further optimization is needed on the basis of confirming the extaction method of Extract Ⅰ. ......
