维药纳气平喘颗粒对哮喘大鼠ICAM1和HO1表达的影响

摘要:目的:研究维药纳气平喘颗粒对哮喘大鼠支气管上皮细胞细胞间粘附分子(ICAM1)和血红素氧合酶1(HO1)表达的影响,以阐明纳气平喘颗粒治疗哮喘病的部分作用机制。方法:将大鼠随机分为正常对照组、哮喘模型组、玉屏风颗粒组、地塞米松组及10.8、5.4、2.7 g/kg纳气平喘颗粒3个剂量组。哮喘模型制备用卵白蛋白(OVA)致敏大鼠并雾化吸入刺激。采用免疫组化法检测各组大鼠支气管上皮细胞ICAM1和HO1表达。结果: 哮喘模型组支气管上皮细胞ICAM1和HO1的表达与正常对照组比较明显增强,10.8、5.4 g/kg纳气平喘颗粒组支气管上皮细胞ICAM1和HO1的表达与模型组相比有不同程度的下降,差异有统计学意义(P<0.01);10.8、5.4 g/kg纳气平喘颗粒组与玉屏风颗粒组和地塞米松组比较差异无统计学意义(P>0.05)。结论:纳气平喘颗粒通过调节支气管上皮细胞ICAM1和HO1的表达,可抑制炎性细胞在气道内聚集及其炎性介质的释放,减少气道变应性炎症,这可能是纳气平喘颗粒治疗哮喘病的部分机制。关键词:纳气平喘颗粒; 哮喘; 细胞间粘附分子1; 血红素氧合酶1
The effect of Ravan Napas on the expression of ICAM1 and HO1 of asthmatic rats

Abstract:Objective:To explore the effect of Ravan Napas on the expression of ICAM1 and HO1 on bronchoepithelial cells of asthmatic rats. Methods:The rats were divided into seven groups randomly: normal control group; asthmatic model group; model group treated with Ravan Napas with three different dosages: high dosage (10.8 g/kg), medium dosage (5.4 g/kg) and small dosage (2.7 g/kg); model group treated with Yu Pin Feng granules; model group treated with dexamethason(DC). Rats were sensitized and challenged with ovalbumin to establish the asthma model. The expression of ICAM1 and HO1 on bronchoepithelial cells were detected through immuno histochemistry method. Results:The expressions of ICAM1 and HO1 on bronchoepithelial cells were significantly higher in asthmatic group than that of the control group (P<0.01). Compared to the asthmatic model group, ICAM1 and HO1 expression was obviously inhibited in the Ravan Napas high and medium dosage treatment groups (P<0.01). There is no significant difference between the Ravan Napas high and medium dosage groups compared to Yu Pin Feng granules group and dexamethason(DC) group (P>0.05). Conclusions: Ravan Napas can reduce the expression of ICAM1 and HO1 on bronchoepithelial cells and thus inhibits the aggregation of the inflammatory cells and the release of the inflammatory media, as a result leads to less inflammation of bronchia. Such inhibitory way might be one of the possible mechanisms of Ravan Napas in treating asthma. ......
