参麦注射液对缺氧复氧致鼠大脑皮层神经细胞Bcl-2/Bax蛋白表达的影响

     【摘要】 目的 研究参麦注射液对缺氧复氧致鼠大脑皮层神经细胞Bcl-2和Bax蛋白表达的影响。方法 取体外原代培养6天的小鼠大脑皮层神经细胞,置于95% N2和 5% CO2的缺氧罐中造成神经细胞不同时间的缺氧复氧。用免疫组织化学法显示缺氧复氧不同时间Bcl-2、Bax蛋白的动态表达,并进行了形态学观察。结果 单纯缺氧复氧组缺氧 8h复氧18h后,可见部分细胞核固缩,同时伴有染色质凝聚及核碎片出现,呈细胞凋亡的典型核形态特征。参麦注射液处理组,缺氧8h复氧18h后,出现核固缩的细胞数目明显减少,大多数细胞核形态正常;缺氧复氧后,大脑皮层神经细胞Bcl-2表达明显减弱,Bax表达明显增强,参麦注射液处理组与单纯缺氧复氧组相比,Bcl-2表达明显增强,Bax表达明显减弱。结论 参麦注射液对小鼠大脑皮层神经细胞缺氧复氧性损伤具有保护作用,而调控Bcl-2 /Bax蛋白表达可能是其重要机制之一。

    【Abstract】 Objective To study protective effects of Shenmai injection on protein Bcl-2 /Bax in mouse cortical neurons injury induced by hypoxia/reoxygenation.Methods The mouse cortical neurons was cultured for 6d, and placed in an anoxia chamber with 95% N2 and 5%CO2 to induce hypoxia/reoxygenation.Phase-contrast microscopy was used to observe morphological characteristics and immunohistochemistry was used to identify neurons.The dynamic expression of Bcl-2 and Bax protein at different H/R time was investigated with immunohistochemical method.Results Nuclei in neurons treated by single H8R18 were partially shrunken and compacted with condensed chromatin. In contrast, the number of it treated by Shenmai injection were reduced,and the most was normal. The expression of protein Bcl-2 decreased while the expression of protein Bax increased following single H/R.After treated by Shenmai injection , the expression of protein Bcl-2 increased while the expression of protein Bax decreased.Conclusion Shenmai injection could effectively protect the nerve cell from injury induced by hypoxia/reoxygenation,and one of mechanism for Shenmai injection could be adjust the expression of protein Bcl-2 and Bax. ......
