关键词: 丝裂霉素C;人结膜囊成纤维细胞;细胞增殖;细胞培养摘 要:目的 观察丝裂霉素C(MMC)对培养人结膜囊成纤维细胞生长及增殖的作用. 方法 用MTT比色法、3 H-TdR掺入法及流式细胞仪检测细胞增殖周期观察MMC对培养的人结膜囊成纤维细胞生长及增殖的影响. 结果 0.4g·L-1 MMC作用5min后,可使成纤维细胞MTT比色A570nm 值7d内无显著变化(P>0.05),3 H-TdR掺入实验CPM第1日较对照组降低了72.5%,至第7日时仍低于对照组86.2%(P<0.01).细胞增殖周期中,MMC组S期细胞的百分比明显低于对照组. 结论 临床使用剂量MMC可使结膜囊成纤维细胞增殖能力下降,处于增殖抑制状态,而不引起明显的细胞死亡.
