【摘要】 目的 探讨大鼠急性肺损伤(ALI)过程中,信号转导和转录活化因子-3(STAT3)的活化规律,及其与细胞因子白介素-6(IL-6)的相关性。方法 健康雌性Wistar大鼠(190±10)g 50只,随机分为生理盐水(NS)组(10只),尾静脉注射NS,2h后处死;LPS模型组(40只),尾静脉注射内毒素(LPS)(5mg/kg),分别于注射LPS后1、2、4、6h时取材。原位杂交法测定肺组织中STAT3的基因表达,放免法检测血和肺泡灌洗液(BALF)中IL-6的浓度,将二者进行相关性分析。结果 急性肺损伤后,在多种肺细胞中均可观察到STAT3的基因表达增加,包括肺泡上皮细胞、支气管上皮细胞、血管内皮细胞以及炎性细胞;正常肺组织STAT3基因弱表达,LPS损伤后肺组织中STAT3的基因表达逐渐增高,4h达峰值,随后其水平逐渐下降,血及BALF中IL-6的浓度变化与STAT3的基因表达相一致,且二者之间明显相关。结论 在急性肺损伤的早期伴随有信号转导JAKs- STAT3通路的激活,IL-6是STAT3通路活化的机制之一。【关键词】 信号转导;转录活化因子-3;急性肺损伤;白细胞介素-6
The expression of STAT3 in acute lung injury rats

【Abstract】 Objective To investigate the activation law of signal transducers and activators of transduction-3(STAT3) and whether cytokine IL-6 can activate STAT3 in acute lung injury (ALI) of rats. Methods 50 female Wistar rats (190±10g) were randomly divided into NS group(n=10), ALI groups(n=40). After intravenous injection of endotoxin (5mg/kg) from tail vein, animals were killed at 1th, 2th, 4th and 6th hour. Detection of STAT3 gene expression of lung tissue with in situ hybridization(ISH) and measurement of the concentration of IL-6 in blood and BALF with radioimmunoassasy.Results LPS treatment resulted in STAT3 activation throughout the resident lung cells, as well as in the recruited inflammatory cells. STAT3mRNA expression in lung constitution increased gradually, peaked at 4th hour and started to decrease at 6th hour. The change of IL-6 concentration in blood and BALF is consistent with the changes of STAT3mRNA expression, and the two is positively correlated. Conclusion The experimental result shows the JAK-STAT3 pathway is activated at early phase of ALI; IL-6 may activate STAT3 pathway at early phase of ALI. ......
