Researchers in Inner Mongolia succeed in preparing

     Han Xiaofang et al the Hospital of Inner Mongolia Autonomous Region in a project entitled " The experimental study of genetically engineered antibody against bacterial endotoxin" successfully prepared human source anti-lipoid antibody (A-Fab), an antagonist of endotoxin. This andtibody is an effective immune agent for the prevention and treatment of endotoxemia. This research project recently won the first grade science and technology award of the Autonomous Region

    Han Xiaofang et al adopted Gram-negativebacilli, J7 endotoxin A protein to immunize healthy volunteer, and then used the technique of antibody pooling by the bacteriophage, successfully established the human source bacteriophage Fab antibody pool. The method of antigen progressive subtraction was used for thrice elutriation and screening of these antibodies to increase the antibody titre to 100 folds of the first elutriation. Then the screening and evaluation of bacteriophage antibody were carried out with the sandwich technique of ELISA; it was discovered that the bacteriophage antibody only conjugated with the lipoid A of endotoxin and had no reaction with the non-correlated antigen, indicating the existence of a specific anti-endotoxin lipoid A in the human source antibody obtained out of screening. By cutting off the gIII gene of bacteriophage, the soluble anti-lipoid A Fab antibody was obtained. Afterwards, Han Xiaofang et al. carried out analysis of heavy chain Fd gene sequence analysis with the two clones of the antibody and discovered that the DNA order of the two clone heavy chain posses the typical feature of gene sequence of human immunoglobulin which belonged to the third kindred member of human antibody., http://www.100md.com
