     Secretory expression of leucinerich repeats of Nogo66 receptor in eukaryotic cells

    【Abstract】 AIM: To construct the eukaryotic secretion vector containing the coding region of human Nogo66 receptor LRR domain and to detect its expression in COS7 cells. METHODS: LRR gene, derived from a prokaryotic plasmid pESHis/LRR, was subcloned into a eukaryotic secretion vector pSectag2B. The recombinant expression plasmid, pSectag2B/LRR, was transfected into COS7 cells with lipofectamine 2000 reagent. RESULTS: Expression of a secreted fusion protein was identified by immunoflurescence assay, Westernblot analysis and sandwich ELISA assay with anticMyc mAb and antiHis antibody. CONCLUSION: Eukaryoric expression vector containing the LRR gene was successfully constructed and expressed in COS7 cells.
