    Culture, observation and identification of neural stem cells

    【Abstract】 AIM: To establish a stable neural stem cell line. METHODS: Fetal mouse striatum from E12 or E16 fetuses were dissected. The tissues were dissociated by digestion and trituration. The dissociated single cells were cultured in DMEM/F12 media containing bFGF (20 mg·L-1), EGF (20 mg·L-1) and N2 supplements. RESULTS: 46 days later, neurospheres were found in cultures, which were redissociated and soon new ones were observed. By immunocytochemistry, the newlyformed neurospheres were found to be nestinpositive and they differentiated into NSEpositive cells and GFAPpositive cells in cultures containing fetal bovine serum. CONCLUSION: It is concluded that cells in these neuropsheres are neural stem cells. By MTT reduction assay, the growth curve of this cell line can be depicted.
