     Effects of potassium antimonyl tartrate on in vitro proliferation and apoptosis of human gastric cancer cell

    【Abstract】 AIM: To explore the therapeutic effects of antimonials on solid tumor by observing the effects of potassium antimonyl tartrate(PAT)on the proliferation and apoptosis of human gastric cancer SGC7901 cells in vitro. METHODS: MTT method was used to measure the levels of the proliferation of SGC7901 cells intervened with PAT of different concentrations. The cell apoptosis was detected by Hoechst 33 258 and flowcytometry. RESULTS: PAT inhibited SGC7901 cell growth significantly in a dose and time dependent manner. PAT displayed prominent inhibitory effects with 20 and 40 μmol/L at 72 hours, and the growth inhibition rates of the cancer cells reached 54.1% and 66.6%, respectively. SGC7901 cells chromatin condensation and nuclear fragmentation were seen under fluorescence microscope in the cells stained with Hoechst 33 258. The apoptosis rate was the highest with 40 μmol/L PAT at 48 hours, reaching 35.2%. Compared with that of the control group, the cell percentage of G0/G1 phase decreased and that of G2/M phase increased (24.6% vs 5.4%). CONCLUSION: PAT significantly inhibits the proliferation and induces the apoptosis of SGC7901 cells. ......
