    Establishment of myocardial stunning model of isolated rat heart

    【Abstract】 AIM: To establish a myocardial stunning model of isolated rat heart by the Langendorff apparatus and identify its reliability. METHODS: 30 rat hearts were infused by KH perfusion solution and were divided to 5 groups by different periods of myocardial ischemia. Myocardial contractile and diastolic functions were measured during experiment and the infarct size was analyzed by TTC staining after reperfusion. RESULTS: There were significant differences in the myocardial function among all the groups during reperfusion (P<0.01) but the differences were not significant before ischemia. The infarct size was (2.2±3.4)%, (7.5±5.2)% and (9.0±4.8)% in the control group, 15 min ischemic group and 20 min ischemic group, respectively (P>0.05). The infarct size was (21.7±5.5)% and (39.7±8.8)% in 25 min ischemic group and 30 min ischemic group, which were significant larger than that in the other three groups (P<0.01). CONCLUSION: The myocardial stunning model established in isolated rat heart is reliable and a 20 min global ischemia is sufficient to induce myocardial stunning. ......
