     DNA fingerprinting of Banna miniature pigs with different probes

    【Abstract】 AIM: To detect the inbreeding degree of Banna miniature pigs with DNA fingerprinting. METHODS: Polymorphic DNA fingerprints from Banna miniature pigs were obtained by Southern blotting with a nonisotopically HRP (Horseradish peroxidase) labelled multiloci minisatellite probe JL02 and 3′HVR and detected by enhanced chemiluminescene. RESULTS: The number of fragments longer than 2 kb with probe JL02 ranged from 13-17. The similarity coefficient of family 805, 807 in substrain JB and family 111, 121, 133, 151 in substrain JS was 0.92±0.04, 0.85±0.13, 0.86±0.02, 0.80±0.05, 0.84±0.09 and 0.88±0.08, respectively, which were significantly higher than those between families (0.46-0.65). The number of fragments longer than 2 kb with probe 3′HVR ranged from 6-15. The similarity coefficient of family 805, 807 in substrain JB and family 111, 121, 133, 151 in substrain JS was 0.94±0.09, 0.85±0.09, 0.84±1.70, 0.78±1.60, 0.85±0.42 and 0.88±0.07, respectively, significantly higher than those between families (0.46-0.65). CONCLUSION: Both probes can be used for DNA fingerprinting of Banna miniature pig, obtaining the same result. The similarity coefficient within families is higher than that between families. The similarity coefficients of family 805 in substrain JB and family 151 in substrain JS are the highest, indicating that the two families have the higher inbreeding degree, smaller genetic difference between individuals and better homogeneity. ......
