【Abstract】 AIM: To investigate the effects of chronic cerebral blood flow shortage caused by blood hyperviscosity on cytosolic free calcium concentration in cerebral cells, MDA and SOD contents in brain tissue. METHODS: Animal model of blood hyperviscosity was established by intravenous injection of high molecular dextran to mice. Alteration of [Ca2+]i in murine cerebral cells was measured by confocal laser scanning microscope using Fluo3/AM as calcium indicator, and MDA and SOD contents in brain tissue were determined. RESULTS: [Ca2+]i in cerebral cells subjected to blood hyperviscosity increased significantly (P<0.01) and [Ca2+]i was positively correlated with the level of blood viscosity (rηH=0.769, rηL=0.821, P<0.01). Compared with those of control, MDA content in the brain tissue rose but the SOD content dropped significantly (P<0.05). The MDA content in the brain tissue was also positively correlated with the level of blood viscosity (rηL=0.720, P<0.01). CONCLUSION: Blood hyperviscosity leads to Ca2+ overload in cerebral cells and the increase of oxygen free radical reaction in brain tissues. ......
