【Abstract】 AIM: To study the induction of MUC1specific CTL in mice by peptide vaccine that was composed of mouse serum albumin and synthetic peptide DMUC1 with partial chirality. METHODS: The DMUC1 peptide containing DSer was synthesized with solid phase peptide synthesis, purified and coupled with mouse serum albumin. BALB/c mice were vaccinated by DMUC1 peptide vaccine and GMCSF or thymopeptide (TP). The abilities to stimulate the proliferation of lymphocytes and to induce the peptidespecific CTL were analyzed by 3HTdR incorporation and release assay, respectively. RESULTS: The molecular weight of the target peptide was confirmed by mass spectrum (MS) analysis. The DMUC1 peptide vaccine stimulated the proliferation of lymphocytes significantly. MUC1specific CTLs could be induced by DMUC1 peptide vaccine and these CTLs could kill the MUC1positive GZ.A5.3H.4 cells. CONCLUSION: The chiral DMUC1 peptide vaccine can induce MUC1specific CTL in mice, which provides some evidence for the treatment of tumors with synthetic DMUC1 peptide vaccine with partial chirality. ......
