     Induction of specific CTLs in BALB/c mice by chiral MUC1 synthetic peptide vaccine

    【Abstract】 AIM: To study the induction of MUC1specific CTL in mice by peptide vaccine that was composed of mouse serum albumin and synthetic peptide DMUC1 with partial chirality. METHODS: The DMUC1 peptide containing DSer was synthesized with solid phase peptide synthesis, purified and coupled with mouse serum albumin. BALB/c mice were vaccinated by DMUC1 peptide vaccine and GMCSF or thymopeptide (TP). The abilities to stimulate the proliferation of lymphocytes and to induce the peptidespecific CTL were analyzed by 3HTdR incorporation and release assay, respectively. RESULTS: The molecular weight of the target peptide was confirmed by mass spectrum (MS) analysis. The DMUC1 peptide vaccine stimulated the proliferation of lymphocytes significantly. MUC1specific CTLs could be induced by DMUC1 peptide vaccine and these CTLs could kill the MUC1positive GZ.A5.3H.4 cells. CONCLUSION: The chiral DMUC1 peptide vaccine can induce MUC1specific CTL in mice, which provides some evidence for the treatment of tumors with synthetic DMUC1 peptide vaccine with partial chirality. ......
