     Study on preparation of 125I-RC160 with NBS and its biodistribution in animals

    【Abstract】AIM: To develop a high efficiency method for directly labeling RC-160 with 125I and to study its biodistribution in normal mice. METHODS: Using Nbromosuccinimide (NBS) as oxidant, rations of RC160 with variable 125I and NBS were added into the reaction system to find the optimal ratio of labeling response. The radioactivity biodistribution was performed in normal mice. RESULTS: The optimal ratio of RC160 ∶125I∶NBS was 10 μg∶7.4 MBq∶30 μg, the labeling efficiency was 76.9% and the radiochemical purity was 95.7% after purification by SepPak C18. 125I-RC-160 was eliminated fast from blood, heart and kidney. It was mainly concentrated in lung, liver, stomach and colon. CONCLUSION: This is an optimal method with simple steps and high labeling efficiency in labeling RC160 with 125I. 125IRC160 is mainly metabolized in digestive system. ......
