【Abstract】AIM: To develop a high efficiency method for directly labeling RC-160 with 125I and to study its biodistribution in normal mice. METHODS: Using Nbromosuccinimide (NBS) as oxidant, rations of RC160 with variable 125I and NBS were added into the reaction system to find the optimal ratio of labeling response. The radioactivity biodistribution was performed in normal mice. RESULTS: The optimal ratio of RC160 ∶125I∶NBS was 10 μg∶7.4 MBq∶30 μg, the labeling efficiency was 76.9% and the radiochemical purity was 95.7% after purification by SepPak C18. 125I-RC-160 was eliminated fast from blood, heart and kidney. It was mainly concentrated in lung, liver, stomach and colon. CONCLUSION: This is an optimal method with simple steps and high labeling efficiency in labeling RC160 with 125I. 125IRC160 is mainly metabolized in digestive system. ......
