Culture of human trophoblast cells from first trimester placentae of human origin

    ¡¾Abstract¡¿AIM£º To prepare highly purified human trophoblast cells from human placentae at the first trimester of normal pregnancy for further study on the function of human placentae villi. METHODS: A standard trypsinª²DNase dispersion method was used to isolate the trophoblast cells and cell purity was determined by immunostaining for markers of fibroblasts (antivimentin). The structural features were revealed by transmission electron microscopy. RESULTS: The cell viability evaluated by trypan blue exclusion was greater than 90%. And light microscopic examination revealed that the cells were epithelialª²like cells, with patchy spreading. The cells were positive for cytokeratin and negative for vimentin, indicating the absence of contaminating elements. Transmission electron microscopy revealed that the cells had fine structural features typical of trophoblasts. CONCLUSION: The method, simple and easy to operate, can be used in studying the function of human placentae villi during pregnancy. ......
