     Role of H2O2 in promoting proliferation of HepG2 cells

    【Abstract】 AIM: To study the role of low concentration of H2O2 in promoting the proliferation of HepG2 cells so as to examine the role of NFκB and cell cycle in that process. METHODS: HepG2 cells were stimulated directly by H2O2. HepG2 cells were previously treated by 20 μmol/L PDTC for 2 hours before they were treated by 50 μmol/L H2O2. HepG2 cells in various phases of cell cycle were treated by 50 μmol/L H2O2. The proliferation of HepG2 cells was detected by MTT assay. RESULTS: H2O2 of 50 μmol/L stimulated the growth of HepG2 cells, while the proliferation induced by H2O2 was prevented by PDTC, an inhibitor of NFκB. H2O2 of 50 μmol/L stimulated the growth of HepG2 cells in G1 phase. CONCLUSION: Low concentration of H2O2 promotes the proliferation of HepG2 cells, partly through a NFκB dependent mechanism. The proliferation of HepG2 cells induced by H2O2 may have different sensibilities in different phases of cell cycle, and it occurs mainly in G1 phase of cell cycle. ......
