     Expression and regulation of intercellular adhesion molecule1 in hepatic oval cells in rats

    【Abstract】 AIM: To investigate the expression of intercellular adhesion molecule1 (ICAM1) in hepatic oval cells (HOCs) and the regulation of nuclear factorκB (NFκB) in the expression of ICAM1. METHODS: Wistar rats were fed on feedstuff with 0.6 g/kg 3′methyldimethylaminoazo benzene(3′MeDAB) for 4 wk, HOCs were isolated by densitygradient centrifugation and immunocytochemistry was performed to detect the expression of ICAM1. HOCs were cultured in vitro respectively with TNFα, activator of NFκB, and TGFβ1, inhibitor of NFκB, and semiquantitative reverse transcriptionpolymerase chain reaction (RTPCR) assay was used to compare the amount of ICAM1 mRNA in HOCs cultured with TNFα and TGFβ1. RESULTS: Immunocytochemistry indicated that ICAM1 expression was positive. Compared with that in control group, the proliferation of HOCs was promoted in TNFα group (P<0.01) while it was influenced nonsignificantly in TGFβ1 group. Semiquantitative RTPCR assay indicated that the expressions of ICAM1 were upregulated in TNFα group (P<0.01) and downregulated in TGFβ1 group (P<0.01). CONCLUSION: ICAM1 is expressed in HOCs and the expression is regulated by NFκB activity change. ......
