关键词: 细胞周期蛋白类;细胞周期蛋白质依赖激酶类;表皮生长因子-尿抑胃素;真皮;成纤维细胞摘 要:目的 探讨表皮生长因子(EGF)对作为皮肤组织工程种子细胞之一的真皮成纤维细胞中cyclinD1和CDK-4表达的影响,以从细胞周期探讨EGF促进真皮成纤维细胞生长的机制. 方法 用含100mL·L-1 胎牛血清的DMEM,加入50mg·L-1 的EGF培养SD大鼠的真皮成纤维细胞,通过MTT检测、流式细胞仪分析观察细胞的生长状态,免疫组化检测cyclinD1和CDK-4的表达,结合流式细胞仪分析观察细胞的周期变化. 结果 MTT检测、流式细胞仪结果都显示50mg·L-1 的EGF能显著促进真皮成纤维细胞的生长增殖,免疫组化结果显示,二者一致. 结论 50mg·L-1 的EGF对真皮成纤维细胞的生长有极大的促进作用,促进了细胞周期蛋白cyclinD1和CDK-4的表达,使细胞G1期变短.

Abstract:AIM To examine the biological behaviors of der-mal fibroblasts in DMEM containing50mg·L-1 EGF and to investigate the expressions of cyclinD1and CDK-4.METH┐ODS Dermal fibroblasts were cultured in DMEM containing50mg·L-1 EGF and100mL·L-1 fetus bovine serum.The proliferation ability was investigated by MTT detection and FCM analysis.The expressions of cyclinD1and CDK-4were investigated by immunohistochemistry method.RESULTS Dermal fibroblasts cultured in DMEM containing50mg·L-1 EGF showed enhanced proliferation and more positive signals for cyclinD1and CDK-4.CONCLUSION The dermal fi-broblasts cultured in DMEM containing50mg·L-1 EGF were in better condition than those cultured in DMEM with-out EGF.And EGF shortened the G1phase of the cell cycle. ......
