谷氨酸单钠对新生小鼠视网膜神经细胞c┐fos表达的影响

关键词: 谷氨酸;兴奋毒性;基因,fos;视网膜;小鼠摘 要:目的 观察谷氨酸对小鼠视网膜神经细胞c-fos表达的变化,探讨谷氨酸的神经毒性作用. 方法 昆明系小鼠妊娠晚期经口一次性大剂量谷氨酸钠ig(4.0g·kg-1 ),仔鼠于出生后10,20和30d取眼球,多聚甲醛固定,恒冷切片,应用免疫组织化学方法对视网膜神经细胞c-fos的表达进行了观测. 结果 10,20和30日龄谷氨酸组小鼠视网膜神经细胞c-fos表达阳性神经元分别为1474,1250及859cells·mm-2 ,比正常小鼠c-fos的表达明显增高(1474vs744cells·mm-2 ,P<0.05;1250vs538cells·mm-2 ,P<0.05;859vs404cells·mm-2 ,P<0.05). 结论 外源性谷氨酸作用于成长发育中仔鼠的视网膜神经细胞,并引起细胞内c-fos过度表达,可能诱导视网膜神经细胞凋亡.
